Summary:
The E26 transformation specific (ETS) transcription factors are involved in gene regulation in all major tissues. As ETS proteins bind DNA via structurally conserved DNA-binding domains, member-specific interactions with DNA are critical determinants of specificity in target gene expression. In cancer, ETS proteins can be oncogenic or tumor suppressive depending on the cellular context. Our group has been interested in the molecular basis of ETS specificity at the protein/DNA level. This talk highlights our recent work on novel mechanisms of specificity for this family of transcription factors and how these insights are enabling promising pharmacologic approaches for inhibiting and enhancing ETS transactivation.
