Unravelling the role of Tubα4A in aging and neurodegenerative diseases

Tubulin α4A is a constituent of microtubules (MTs), a dynamic structure involved in several biological processes. This tubulin α isoform was shown to be expressed according to aging. Mutations of this protein are also associated with a neurodegenerative disease, the amyotrophic lateral sclerosis (ALS). In addition, it was demonstrated that Tubα4A is kept in a de-tyrosinated state, which could confer a higher stability to the associated MT. Despite those discoveries, little is known about its relationship with aging and its physiological role.
By using a transgenic mouse model expressing an HA-tagged version of Tubα4A, we aim to discover the expression profile of this protein throughout aging. We are also currently using motor neurons differentiated from human stem cells to study Tubα4A function.
