Talk Title: Deciphering human organogenesis principles with single-cell and spatial technologies

Biography

Dr. Peng He is a postdoctoral researcher at the European Bioinformatics Institute jointly supervised by Dr. John Marioni and Dr. Sarah Teichmann. Dr. He holds a BSc in Statistics and Biochemistry from the University of Hong Kong, and a Ph.D. in genetics from California Institute of Technology. Dr. He worked in mouse organogenesis and high-throughput sequencing data analysis as part of the ENCODE project for his thesis. His current research projects focus on human developmental processes of the embryonic limb and the fetal lung using single-cell and spatial methods.

Abstract

Human embryo development involves sophisticated cell-fate determination and execution. However, the existing references of cell types and cell states are still incomplete due to limitations in sampling and analytic methods. Here I present a systematic approach combining single-cell transcriptomics and accessibility, spatial transcriptomics, and novel computational methods to show an updated view of human tissue development, using the limb and the lung as test cases. We identified novel epithelial subtypes including a cancer-related lineage in the lung and new anatomical structures in the distal limb. We further reproduced the transcriptomic phenotypes of the neuroendocrine lineage in the lung NE cell organoid system. Our annotated datasets and workflow will provide valuable resources for the community of developmental biologists.
