Summary:
Pluripotent embryonic stem cells (ESCs) have a unique cell cycle and gene expression program that facilitates lineage determination alongside cellular scaling during development. Dr. Natarajan’s team generate FUCCI-mESCs that capture spatio-temporal cell cycle progression at the singlecell level. Here Dr. Natarajan presents their work investigating cell cycle dynamics combining flow cytometry (FACS) and RNA sequencing (scRNA-seq) of ~700 single ESCs throughout entire cell cycle and across two culture conditions. They integrate single-cell measurements and assess how cellular features affect gene dynamics during cell cycle, reconstruct single-cell gene expression profiles and predict gene contribution to cell cycle, stem cell fate and in maintaining cellular homeostasis.
