Summary:
The synthesis of new ribosomes is a fundamental conserved process required for protein synthesis in all cells. Mutations in key ribosome assembly factors characterise the ribosomopathies, a growing new class of cancer predisposition disorders. Ribosomal subunits pre-assembled in the nucleus are exported to the cytoplasm where they acquire functionality in a final series of maturation steps that result in formation of the catalytic core of the ribosome. Using a combination of genetic, biochemical and structural approaches, Professor Warren's team's work has provided new insights into how the central catalytic site of the ribosome is sculpted, how this process is monitored and how it is disrupted in leukaemia-associated ribosomopathies.
