Biography

Dr. Katsuyuki Shiroguchi is a Team Director at the RIKEN Center for Biosystems Dynamics Research (BDR), Japan. He received his PhD from the University of Tokyo, where he began his research career in biophysics studying the mechanism of molecular motors by observing single molecule dynamics under a microscope. Later during his postdoctoral training at Harvard University, he switched to genomics and developed digital RNA sequencing using originally developed DNA molecular barcodes. His current research at RIKEN focuses on single cell analysis to better understand multicellular systems by combining imaging, sequencing, machine learning, and robotics. His targets include stem cells, organoids, immune system, and cancer.

Abstract

Biological systems are often complicated, and the integration of various modalities in measurements is a powerful approach for understanding such biological systems. We are integrating an optical microscope, our developed Automated Live imaging and cell Picking System (ALPS), transcriptomics, and image-based deep learning. This integration enables us to estimate the gene expression states of individual cultured cells in Epithelial Mesenchymal Transition, to construct a platform for finding markers of organoid-forming cells from multiple tissues, and to develop a new method of cell-based spatial omics for three-dimensional samples. In this talk, I will share these approaches, some biological results, and future possibilities.
